Yes, you can set the preloads with the diff on the truck. It's way easier to do it with the diff out though. Setting backlash would be a royal bitch with the diff on the truck.
You could pull the diff out in 3 pieces, reassemble/torque it outside the truck, set all your critical measurements, and then disassemble and reinstall that way... All the measurements would hold as long as you properly torqued the case bolts once in the truck.
