I can't tell you from experience, but it depends on what type of locker you're talking about. If you're just talking lunchbox lockers (lockright, detroit, etc), they don't replace the existing carrier, but rather go inside of it. So as long as your diff is an "open" carrier, you can just stick a lunchbox locker in there and thats it. With an Elocker, air locker, and other carrier type lockers, they replace the entire carrier so you basically have to disassemble the entire diff/axle and rebuild it from the ground up. That includes bearing, seals, shimming, gear meshing, backlash, all that technical stuff. I don't quite trust myself to do the work so I'm just taking all of this stuff to a guy at a transmission shop and letting him have all of the fun.
